(2019) ‘Leaky’ Vaccines Can Produce

https://www.healthline.com/health-news/leak...-viruses-072715

Article written April 5, 2019

The current debate over vaccinating toddlers is small potatoes compared to the potential risks of using “leaky” vaccines to prevent disease.

According to a new study published today in the scientific journal PLOS Biology, some types of vaccines could allow more virulent versions of a virus to survive, putting those who are unvaccinated at greater risk of severe illness.

To understand this, it’s necessary to examine the difference between “perfect” vaccines and “leaky” ones.

Perfect vaccines are so-named because they mimic the perfect immunity that humans naturally develop after having certain childhood disease.

“When a vaccine works perfectly, as do the childhood vaccines for smallpox, polio, mumps, rubella and measles, it prevents vaccinated individuals from being sickened by the disease, and it also prevents them from transmitting the virus to others,” said Andrew Read, an author of the study and an Evan Pugh professor of biology and entomology and Eberly professor in biotechnology at Penn State University.

What We Learned from Chickens

Enter the chickens.

Read was studying vaccines that could combat malaria when he got interested in Marek’s disease, a highly contagious viral disease that affects chickens.

It’s a form of herpes that is found in chicken dander and is more virulent than the Ebola virus, Read said.

After experiments done in a specialized pathogen-containment facility at The Pirbright Institute in the United Kingdom, the researchers concluded that the vaccines developed to combat Marek’s disease were imperfect or leaky.

“These vaccines also allow the virulent virus to continue evolving precisely because they allow the vaccinated individuals, and therefore themselves, to survive,” said Venugopal Nair, who led the research team. He is the head of the Avian Viral Diseases program at The Pirbright Institute.

These less-than-perfect vaccines create a “leaky” barrier against the virus. Vaccinated individuals may get sick but have less severe symptoms, but the virus survives long enough to transmit to others, which allows it to survive and spread throughout a population.

“Our research demonstrates that the use of leaky vaccines can promote the evolution of nastier ‘hot’ viral strains that put unvaccinated individuals at greater risk,” Nair said.

Marek’s disease used to be a minor ailment that did little harm to chickens in the 1950s, but the virus has grown stronger and today is capable of killing all the unvaccinated birds in poultry flocks, sometimes within 10 days.

But since nearly every chicken in agricultural production throughout the world is vaccinated, Marek’s is a relatively minor problem today.

Preventing More-Virulent Virus Strains

Marek’s is not the only nasty disease out there. The virus causing avian influenza can be even deadlier.

“The most virulent strain of avian influenza now decimating poultry flocks worldwide can kill unvaccinated birds in just under three days,” Read said, because the vaccine against avian influenza is a leaky one.

“In the United States and Europe, the birds that get avian influenza are culled, so no further evolution of the virus is possible,” Read said.

Culling is a more expensive process than using a leaky vaccine, he said. But it’s safer.

“Instead of controlling the disease by culling infected birds, farmers in Southeast Asia use vaccines that leak — so evolution of the avian influenza virus toward greater virulence could happen,” he added.

Some human deaths from avian influenza virus have been reported in China.

“We now are entering an era when we are starting to develop next-generation vaccines that are ‘leaky’ because they are for diseases that do not do a good job of producing strong natural immunity — diseases like HIV and malaria,” Read said.

What’s the answer?

Rigorous testing and vigilant monitoring of next-generation vaccines to prevent the evolution of more-virulent strains of viruses will help.

Read sees this as crucial to the current attempt to develop an Ebola vaccine. He notes that secondary techniques can help when using leaky vaccines, such as insecticide-treated bed nets for prevention of malaria.

A promising new treatment for COVID-19 infection

A flowering plant native to North Africa and Western Asia could be utilised in the future treatment of COVID-19 infection.

The seeds of the plant, Nigella Sativa, have been used for centuries as a traditional remedy for multiple medical conditions, including inflammation and infections. Now, an Australian-first research review article has found it could be used to treat COVID-19.

“There is growing evidence from modelling studies that thymoquinone, an active ingredient of Nigella Sativa, more commonly known as the Fennel Flower, can stick to the COVID- 19 virus spike protein and stop the virus from causing a lung infection.

“It may also block the ‘cytokine’ storm that affects seriously ill patients who are hospitalised with COVID-19,” said Professor Kaneez Fatima Shad, lead author of a recently published comprehensive review article in the prestigious journal, Clinical and Experimental Pharmacology and Physiology.

Thymoquinone has been extensively studied in laboratories, including animal studies. These studies have shown that thymoquinone can moderate our immune system in a good way, by preventing pro-inflammation chemicals such as interleukins from been released.

This gives thymoquinone a potential role as a treatment for allergic conditions such as asthma, eczema, arthritis conditions including rheumatoid and osteoarthritis and even possibly multiple sclerosis.

The review paper details the mechanisms of action of Nigella Sativa and thymoquinone and how they are a promising future treatment of COVID-19 infection. There have been many barriers to the development of Nigella Sativa as a therapeutic agent in large part due to its poor natural gastrointestinal absorption.

“Advances in pharmacological development such as nanotechnology have seen the chance to overcome this barrier to enable for its use as an effective oral medication.

“Furthermore, the drug has recently been successfully given to patients as a nasal spray and topical paste,” said Dr Wissam Soubra, co-author.

Nigella Sativa has been shown to be helpful in treating high blood pressure, high cholesterol and diabetes mellitus. As an anti-inflammatory treatment, Nigella Sativa has also been found to help patients with allergic rhinitis and sinusitis, eczema, osteoarthritis and childhood epilepsy.

Nigella Sativa has also been proven to be effective in a laboratory environment in killing bacteria such as staphylococcus aureus that can cause a range of mild to severe infections if they enter the skin, and viruses including influenza.

“The review paper provides insight into a natural product that has been used as a traditional remedy for over thousand years and may be finally receiving the recognition it deserves,” said Associate Professor Dennis Cordato, co-author.

The study, The role of thymoquinone, a major constituent of Nigella sativa, in the treatment of inflammatory and infectious diseases , was recently published in the journal Clinical and Experimental Pharmacology and Physiology.

https://www.uts.edu.au/news/health-science/...id-19-infection

Why ivermectin? why no love for

I've been looking supplements to improve our immune system. Other than vitamin C and D3, I found this study done by few on HNS (honey + nigella sativa) not just boost immune system but have anti-inflammation and anti-viral effect. People focused on drugs, why not natural resources for therapeutic treatment? Sotrovimab costs USD2200, ivermectin not safe if overdose.
What motorcycle faster than car

Hi,

Looking to upgrade my FZ150i with another more powerful bikes as I'm commuting to work daily using non-highway, NARROW back road. The problem is always gets bullied by cars and modded rempit mopeds.

The mopeds I dont mind, but the cars always overtake and squeezing into my lane, despite following closely car in front.

I want to overtake, so I'm utilizing the rpm limit but without any significant acceleration . The road only has 1 lane for each direction, so its quite dangerous for slow bikes to overtake cars.

Do you think 250cc makes a huge difference? or do i need to look into B full bikes like 650cc? Thanks sifu!

Intel Graduate prgrm vs current work

Hi, as title above... Can I have your opinions about the Intel Graduate Trainee Program regarding the future career opportunity? (eg: permanent employement)

I'm currently a fresh graduate who just got employed in a electronics manufacturing factory as Assistant Engineer about 1 month ago, with 1 year contract as probation. The company is really near to my home in sp....about 5 km. My starting gross pay is RM2k+.

I got a call from Intel penang/kulim asking whether if I'm interested in the program; hence will be notified if I was shortlisted for the interview. I ask what will be the job scope, she said depends later on....after I got accepted.

Should I stick with current company, gaining enough exp for 1-2 years before trying into other larger companies?
Hope you guys can give opinion about my situation...Thanks!
fz150i (new/used) vs pulsar ns200 new ?

Hi guys, based on title, whats best for me if the usage is 70/30 (city-highway)?

My current workplace distance is about 10km from my home, therefore i feel bigger size bikes would be awkward for daily commute. However, I would like to travel long distance (kedah -penang,perak,kl) once in a while like weekends or public holidays.

But the price of new fz150i vs modenas pulsar ns200 is not much of a difference.

Hope to hear your opinions!

Fresh Graduate no prior exp?

Hi guys, as title...

I'm a fresh graduate of bachelor degree engineering.... But I have very little experience in industry. I've done a compulsory internship for 2 months during uni, working as the technician assistant in a small company that's not related to my study.

So now I've completed my study, can you guys suggest me what's the best for career growth?

Should I seek internship/graduate training instead of full time positions (entry level) which the fresh graduates are encouraged to apply?

ex-Melaka coach - MY football will never progress

https://www.fourfourtwo.com/my/features/for...-never-progress

QUOTE

Former Melaka coach Eric Williams has made startling revelations over the Malaysia Super League (MSL) side’s unprofessional conduct in all aspects from management to players, and how these issues contributed to his eventual sacking.
The Englishman, who carries an Australian passport, believes he was victimised throughout the six months at Melaka since his arrival in November last year.

He branded some of the local players, who are highly paid, as ‘lazy’ and also questioned the team’s football agent for recommending ‘average’ players.

Williams described team manager Datuk Yusoff Mahadi as a fickle-minded person who listened too much to his close associates, that included a greedy football agent.
The former Yangon United trainer left for good recently after signing his termination letter on July 6, about seven weeks after he was rested by the Melaka United Soccer Association (MUSA) for the team’s poor form.

He described his short spell with Melaka as something he had never encountered before throughout his coaching career that spans over 29 years.
Prior to joining the MSL, the 61-year-old had worked in Australia, Indonesia, Maldives and Myanmar.
“I had never encountered a situation where players can dictate terms with the management. They wanted me out,” said Williams.
“If this unhealthy trend continues within the clubs in the MSL, Malaysian football will never progress.”
Williams narrated to FourFourTwo on matters that led to his sacking.

On June 2, my contract was terminated. The reason…a group of players met Yusoff the previous month and voiced their dissatisfaction over my training methods.
“They claimed my training was too hard…basically they disliked the intensity of my practice sessions.
“Frankly, it was tough with these players. They would request for unrecorded days off and were disinterested in doing daily double training sessions. They did not like morning sessions and would often produce medical certificates to cover their lies.
“They felt I was challenging their commitment and intelligence.
“These players had no ambition and no desire to be better in the sport. They were only interested in getting their salaries, but were reluctant to adapt with new training methods.
“I blame the agent and the technical director for signing this group of inexperienced players from lower teams”.

Read more at https://www.fourfourtwo.com/my/features/for...c9LQ5hRfpoup.99

Is it the same as in white collar industry in malaysia?
